Detection and mass spectrometry identification of protein changes in low-abundance tissue using CyDye DIGE Fluor saturation dyes

چکیده
Traditional two-dimensional electrophoresis (2-DE) is a wellestablished technique for protein analysis, but its use in identifying changes in protein abundance in tissue samples has been limited by system variability and low sensitivity. These limitations have largely precluded its use to identify protein differences in scarce materials such as those recovered from laser-capture microdissection (LCM).

Selective Labelling of Cell-surface Proteins using CyDye DIGE Fluor Minimal Dyes
Surface proteins are central to the cell's ability to react to its environment and to interact with neighboring cells. They are known to be inducers of almost all intracellular signaling. Moreover, they play an important role in environmental adaptation and drug treatment, and are often involved in disease pathogenesis and pathology (1). Protein-protein interactions are intrinsic to signaling p...
متن کاملIdentification of crystallin modifications in the human lens cortex and nucleus using laser capture microdissection and CyDye labeling
PURPOSE With aging, lens crystallins undergo post-translational modifications (PTMs) and these modifications are believed to play a major role in age-related cataract development. The purpose of the present study was to determine the protein profiles of crystallins and their PTMs in the cortical and nuclear regions within an aging human lens to gain a better understanding about changes in cryst...
متن کاملProteomic Analysis of Bovine Pregnancy-specific Serum Proteins by 2D Fluorescence Difference Gel Electrophoresis
Two dimensional-fluorescence difference gel electrophoresis (2D DIGE) is an emerging technique for comparative proteomics, which improves the reproducibility and reliability of differential protein expression analysis between samples. The purpose of this study was to investigate bovine pregnancy-specific proteins in the proteome between bovine pregnant and non-pregnant serum using DIGE techniqu...
متن کاملProteomic analysis of muscle tissue from rainbow trout (Oncorhynchus mykiss) fed dietary β-glucan
The aim of this study was to examine the changes in muscle proteome of the rainbow trout fed dietary β-glucan. The experimental diets contained 0 (control), 0.1% and 0.2% β-1,3/1,6 yeast glucan. First, feeding larvae were fed to apparent satiation nine times per day with their respective diets over two months. The percentage of body weight gain and feed efficiency of fish fed 0.2% diet was sign...
متن کاملDifferential proteomic screening and identification for non-traumatic necrotic femoral osseous tissue
Currently, there is a lack of effective early screening and detection methods for femoral head necrosis. Current research on most orthopedic diseases focuses on proteomics in the preliminary stage. The recent fluorescence differential in gel electrophoresis (DIGE) has advantages such as a high reproducibility, high sensitivity, high throughput, and high dynamic range. It is currently one of the...
